Interleukin-6, interferon-γ, interleukin-8, and granulocyte-macrophage colony stimulating factor levels in human amniotic fluid at term

Abstract

Cytokines contribute to the maintenance of successful pregnancy and have also been implicated in the initiation of labour. This study has examined those cytokines which may be involved in normal term parturition. IL-6, IFN-γ, IL-8 and GM-CSF levels were measured by ELISA in samples of amniotic fluid, peripheral and uterine venous blood plasma, and umbilical artery blood plasma from non-labouring women undergoing elective caesarean section at term and from women in spontaneous labour. IFN-γ and IL-6 were detected only in amniotic fluid and not in blood plasma samples, unlike IL-8 which was found in all amniotic fluid and plasma samples from both labouring and non-labouring women; GM-CSF was undetectable in all samples examined. Levels of IL-6 were significantly raised in the amniotic fluid of women in labour compared with the control group (P = 0.008). IFN-γ was detected in the amniotic fluid of both labouring women and the control group, and there was also a significant correlation (P = 0.003) between IFN-γ and IL-6 levels in individual amniotic fluid samples. The presence of both IFN-γ and IL-8 in amniotic fluid, as well as IL-6, suggests these cytokines are involved in important immunobiological events relevant to the latter half of gestation.

      Interestingly, it has been shown that preterm infants overexpressed genes that are involved in the negative regulation of IFN-γ production (Olin et al., 2018). Moreover, IFN-γ was detected in AF of both labouring women and elective CS at term (Olah et al., 1996), suggesting that this cytokine is important in pregnancy and delivery but not at all indicative of inflammation related to premature delivery. IFN-γ increase as pregnancy progresses and is elevated around term and therefore decreased in AF samples of the PTB patients included in our study compared to TD.

      Expression levels of inflammatory cytokines from trophoblasts and other cells in the normal placenta were previously reported (Kameda et al., 1990; Kauma et al., 1993). IL-6 levels in maternal serum (Arntzen et al., 1997), amniotic fluid (Olah et al., 1996), vaginal fluid (Imseis et al., 1997) and the placenta (Steinborn et al., 1996) were found to increase during the process of normal labor relative to the non-labor state. Previous work indicating a significant positive correlation between MT gene expression and IL-6 suggests that increased IL-6 expression is strictly related to an enhanced production of MTs (Giacconi et al., 2007).
